Medical Description

Trikonol Plus tablet is a prescription medicine used to treat depression and depression-associated anxiety. Depression is a mental health disorder characterised by persistent depressed mood or a loss of interest in activities for most of the day, every day. Poor focus, feelings of excessive guilt or low self-worth, hopelessness about the future, thoughts of death or suicide, disrupted sleep, changes in appetite, and feeling especially weary or lacking in energy are all possible symptoms. The active ingredients in Trikonol Plus tablets are Amitriptyline and Chlordiazepoxide. Amitriptyline belongs to a class of medicines called tricyclic antidepressants, and Chlordiazepoxide is a benzodiazepine. Directions for use of Trikonol Plus tablets should be strictly followed. Sudden stopping of this medication is not advised. It is important to inform the treating doctor of all medicines taken so that they can be checked for possible interaction issues. Side effects of Trikonol Plus tablets include dry mouth, nausea, vomiting, constipation, tiredness, weakness, nightmares, headache, dry mouth, difficulty urinating, blurred vision, burning sensations or tingling in the hands or feet, excessive perspiration, changes in sexual desire, changes in appetite or weight, disorientation, and unsteadiness.

  • Trikonol Plus tablet is used to treat Major Depressive Disorder associated with moderate to severe anxiety.

Contraindications

Do not take Trikonol Plus tablets or use caution: If you have hypersensitivity reactions to the active or inactive components of this medicine If you have a history of heart conditions like irregular heart rhythm and heart block or had a myocardial infarction (heart attack) recently If you are on treatment with monoamine oxidase inhibitors (MAOIs) like selegiline If you have a severe liver impairment or disease If you have severe lung disease or lung failure

Trikonol Plus tablets are not indicated in pregnant women. There is limited clinical data about its use in pregnant women. Animal studies have shown reproductive toxicity. Consult your physician.

Driving

Alcohol

Talk to your doctor if
  • You have liver disease.

  • You are an older adult. You may be more sensitive to the side effects of antidepressants.
  • Your test reports show changes in liver function after starting Trikonol Plus tablets.
  • You develop headaches associated with eye pain.
  • You develop or have an episode of blood vomiting.
  • You develop abnormal limb movements after starting treatment with Trikonol Plus tablets.
  • You have seizures for the first time after taking Trikonol Plus tablets.

Directions for Use

  • Take Trikonol Plus tablets exactly as advised by your doctor.
  • Do not exceed the dosage, even if you cannot notice the effects of this medicine.
  • Take Trikonol Plus tablets as a whole with a glass of water; do not break or chew this tablet.
  • Take this medicine with or without food.
  • Do not take Trikonol Plus tablets with other medications without informing your doctor.
  • Do not abruptly stop taking Trikonol Plus tablets; this can result in adverse effects from withdrawal.

Storage and disposal

Store Trikonol Plus tablets in a cool and dry place, away from direct sunlight and humidity. Keep away from the reach of children and pets. Dispose of the unused medicine carefully. Do not consume Trikonol Plus tablets if the package was already opened or beyond the expiry date.

Overdose

Trikonol Plus tablet overdose may cause urinary retention, lowered consciousness, seizure, coma, increased or irregular heartbeat, low blood pressure, heart block, dilated pupils, dry mouth, and respiratory depression. In case of a suspected overdose, immediately rush the affected person to the nearest hospital for emergency management.

Missed a Dose

If you have missed a dose of the Trikonol Plus tablet, try to take the missed dose immediately. But if it is already time to take the next dose, skip the missed dose and take the next one. Do not take both doses together or within a few hours as it can cause an overdose.

Mode of Action

How Does It Work?

Trikonol Plus tablet is a combination of two medicines: Amitriptyline and Chlordiazepoxide. Amitriptyline belongs to a class of medications called tricyclic antidepressants (TCAs), which act on multiple brain pathways to increase levels of certain chemical messengers called neurotransmitters. It blocks the membrane pump mechanism that allows neurotransmitters like norepinephrine and serotonin to re-enter the brain, increasing their concentration in synaptic clefts. It also causes sleepiness. In the long term, it increases the levels of brain-derived neurotrophic factor (BDNF), which reduces the symptoms of Major Depressive Disorder. Chlordiazepoxide is a benzodiazepine. It produces sedating and calming effects by acting on the brain to increase the effects of a neurotransmitter, gamma-aminobutyric acid (GABA).

Interactions with other medicines

  • Trikonol Plus tablets are known to interact with the following medications:
  • Medicines that affect heart rhythm like (cisapride and astemizole) can result in potentially life-threatening conditions like heart block or cardiac arrest.
  • Monoamine oxidase inhibitors (MAOIs) like selegiline and rasagiline increase the risk of serotonin syndrome (high serotonin levels in the body causing symptoms like confusion, hallucinations, seizures, extreme changes in blood pressure, blurry vision, and excessive sweating).
  • There is an increase in the level of central nervous system depressants like alprazolam when co-administered with Trikonol Plus tablets. This can cause increased drowsiness.
  • Antihypertensive medications like propranolol and metoprolol reduce the metabolism of Trikonol Plus tablets, causing an increased risk of side effects.
  • Anti-anxiety medication like escitalopram reduces the metabolism of Trikonol Plus tablets, causing an increased risk of side effects.
  • Anti-allergic medications like cetirizine increase the central nervous system depressant (CNS depressant) activities of Trikonol Plus tablets.
  • Painkillers like diclofenac increase the risk or severity of gastrointestinal bleeding.
  • Antifungal medications like ketoconazole decrease the metabolism of Trikonol Plus tablets, causing an increased risk of side effects.
  • Blood thinners and antiplatelet agents like warfarin and aspirin increase the risk or severity of gastrointestinal bleeding.
  • Migraine medicines like zolmitriptan and sumatriptan increase the central nervous system depressant (CNS depressant) activities of Trikonol Plus tablets.
  • Antiviral medicines used to treat HIV/AIDs like nelfinavir, ritonavir, saquinavir, and indinavir decrease the metabolism of Trikonol Plus tablets, causing an increased risk of side effects.
  • Antibiotics like clarithromycin may increase the blood levels of Trikonol Plus tablets, leading to an increased risk of side effects.

Frequently Asked Questions (FAQs)

People with liver disease should exercise caution if using Trikonol Plus tablets. The doctor will decide on dosage adjustments. Regular monitoring of liver function tests (LFT) is generally recommended.

Trikonol Plus tablets can have undesirable side effects like lowering sexual desire (libido), erectile dysfunction, and an inability to reach orgasm. If you experience these problems, discuss them with your doctor.

Consult your doctor before taking Trikonol Plus tablets if you have diabetes, as it might fluctuate blood glucose levels. Regular monitoring of blood sugar levels is advised. Talk to your doctor if you experience fluctuations in blood sugar levels after starting treatment.
