Medical Description

Amitone Forte is a prescription medication used in the treatment of depression and associated anxiety. It is a combination of two drugs, Amitriptyline (25 mg) and Chlordiazepoxide (10 mg). Amitriptyline belongs to a class of medicines called tricyclic antidepressants, and Chlordiazepoxide is a benzodiazepine. The mode of action of Amitone Forte is by affecting multiple brain pathways that regulate mood, personality, and anxiety by targeting chemical messengers called neurotransmitters; hence their use as antidepressants. Depression is a mood disorder characterised by low mood, loss of interest in daily activities, difficulty sleeping, and other features. Anxiety is a condition associated with nervousness and constant thoughts of worry. These conditions are related to altered levels of neurotransmitters in the brain. Directions for use Amitone Forte should be strictly followed. The medication should be taken only after consulting a doctor. It should not be abruptly started or stopped. Taking the tablet at the same time every day is advisable. Side effects of Amitone Forte include constipation, urinary retention, dry mouth, orthostatic hypotension and dizziness, blurred vision, confusion, and increased appetite and weight gain.

Uses

  • Amitone Forte is used for the treatment of Major Depressive Disorder (MDD) associated with moderate to severe anxiety.

Contraindications

  • Do not use Amitone Forte or use with caution:
  • If you have hypersensitivity reactions to the active or inactive components of this medicine.
  • If you have a history of heart conditions such as irregular heart rhythm or had a myocardial infarction (heart attack) recently.
  • If you have an elective surgery planned; stop this medication before your surgery to avoid the risk of interaction with anaesthetic agents.
  • If you have angle-closure glaucoma; do not use this medicine.
  • If you have any epilepsy/seizure disorders; this medication decreases the seizure threshold; use cautiously.
  • If you have any pulmonary diseases; use this medicine cautiously.
  • If you are pregnant or planning a pregnancy.
  • If you are under 24 years of age; this medication may increase suicidal ideation in this age group.

Overdose

Overdose of Amitone Forte may cause sedation, seizure, coma, increased heart rate, low blood pressure, dilated pupils, dry mouth, and difficulty in passing urine or stools. In case of a suspected overdose, immediately rush the affected person to the nearest hospital for emergency management as an over can is a life-threatening condition.

Missed a Dose

If you have missed a single dose of the Amitone Forte tablet, take it as soon as you remember. If you recall missing a dose when it's almost the time to take the next dose, skip the missed dose and continue with your regular dosing schedule. Do not take both of them together, as this can lead to an overdose. This tablet should be used only when needed or as prescribed by the doctor. Talk to your doctor if you have any confusion about the dosage.

How Does It Work?

Amitriptyline belongs to a class of medications called tricyclic antidepressants (TCAs), which act on multiple brain pathways to increase levels of certain chemical messengers called neurotransmitters. It works by increasing the levels of neurotransmitters like serotonin and nor-adrenaline that affect mood and personality. It also causes sleepiness. In the long term, it increases the levels of BDNF (brain-derived neurotrophic factor), which reduces the symptoms of Major Depressive Disorder. Chlordiazepoxide belongs to a class of medications called benzodiazepines. It produces sedating and calming effects in users by acting on the brain to increase the effects of a neurotransmitter called GABA (gamma-aminobutyric acid).

Interactions

Interactions with other medicines

  • This medication affects the heart rhythm. It should not be used with other medications like astemizole and cisapride that also have the same effect. This may result in potentially life-threatening conditions.
  • This medication should not be used concomitantly with MAOIs (Monoamine oxidase inhibitors) or within 14 days of the use of MAOIs and vice versa. Examples of MAOIs include selegiline and rasagiline.
  • This medication should not be used with other medicines like isocarboxazid, which increases serotonin levels. This may result in a potentially fatal condition.
  • This medication should not be used with other CNS depressants.
  • This medication should not be mixed with other benzodiazepines.

Amitone Forte can take 4 to 6 weeks for efficacious pharmacological action. Do not increase the dose without the express prescription of your treating doctor.
