Medical Description
Betatinib capsule is an anticancer medicine containing acalabrutinib as its active ingredient. It belongs to a group of medicines called Bruton's tyrosine kinase (BTK) inhibitors. It is used to treat certain blood cancers, including mantle cell lymphoma (MCL), chronic lymphocytic leukaemia (CLL), and small lymphocytic lymphoma (SLL) . These cancers affect lymphocytes, a type of white blood cell that plays an important role in the immune system.
Acalabrutinib works by binding to and inhibiting the activity of BTK, a protein involved in signalling pathways that help certain B cells grow, survive, and function.. . By inhibiting BTK signalling, Betatinib capsule interferes with pathways involved in the growth and survival of abnormal B cells.
Before starting treatment, tell your doctor if you have bleeding problems, an infection, heart rhythm problems, liver problems, or if you have recently had or are planning surgery. Also, inform your doctor if you are pregnant, planning a pregnancy, or breastfeeding.
Betatinib capsule may increase the risk of bleeding, infections, low blood cell counts, certain second primary cancers, and abnormal heart rhythms such as atrial fibrillation or flutter. . Your doctor may carry out regular blood tests and other checks during treatment. Tell your doctor about all medicines you take, especially blood-thinning medicines, as they may further increase bleeding risk.
Betatinib capsule is not generally recommended for children and adolescents because its safety and effectiveness in this age group have not been established. Take this medicine only as prescribed by a doctor experienced in treating blood cancers.

Side effects
- Low red blood cell count (anaemia), causing tiredness and breathlessness
- Low neutrophil count (neutropenia), which may increase the risk of infections
- Infections of the nose, throat, or upper airways (like the common cold or sore throat)
- Low platelet count, causing easy bruising or bleeding
- Headache
- Diarrhoea
- Muscle and joint pain
- Tiredness and weakness
- Skin rash
- Feeling sick (nausea) or vomiting
- Constipation
- Dizziness
- High blood pressure
- Raised liver readings on blood tests
- Infections caused by the herpes virus
- Development of certain second primary cancers, including skin cancers
- Serious or life-threatening infections, including lung infection (pneumonia), blood infection (sepsis), and rare infections that can affect the lungs or brain
- Serious or life-threatening internal bleeding, including in the brain or gut
- Very low blood counts that may require treatment interruption, dose adjustment, or other medical treatment
- Irregular heartbeat, including a condition called atrial fibrillation, causing fluttering in the chest, dizziness, or discomfort

- You notice unusual bleeding, such as blood in your stools, pink or brown urine, long nosebleeds, or unexpected bruising, as this medicine can cause serious bleeding, especially if you also take blood-thinning medicines or aspirin-type medicines.
- You develop fever, a persistent cough, chills, or any signs of infection, as this medicine can lower certain white blood cell counts and increase your risk of serious infections, including lung, blood, and opportunistic infections.
- You have had a hepatitis B infection in the past, as this medicine can cause it to become active again; your doctor may check for hepatitis B infection and determine whether monitoring or preventive treatment is needed.
- You feel your heart beating unevenly, too fast, fluttering in the chest, feel faint, short of breath, or have chest discomfort, as this medicine can sometimes cause an irregular heartbeat, especially in those who already have heart conditions or high blood pressure.
- Watch for any new, growing, or unusual skin spots or marks, as this medicine is linked to a higher risk of certain second primary cancers, including skin cancers; wear sun protection and get your skin checked regularly.
- You are planning surgery or any dental work, as you may need to pause this medicine for 3 to 7 days before and after the procedure to lower the risk of bleeding.
- You have severe liver disease, as this medicine should not be taken in this situation.
- You take any indigestion medicines, including PPIs, H2-blockers, or antacids, as some can reduce your body's absorption of this medicine and may require dose adjustments.
- You take blood thinners or aspirin-type medicines, as combining these with Betatinib capsule can raise your risk of serious bleeding.
- You are thinking about having children in the future, as you should discuss reproductive considerations with your doctor.
Directions for Use
- Take Betatinib capsule exactly as your doctor tells you, with or without food.
- Swallow the capsule whole with a full glass of water; do not open, break, or chew them.
- If you take antacids, take them either 2 hours before or 2 hours after Betatinib capsule; if you take a stomach acid medicine called an H2-blocker (such as famotidine), take Betatinib capsule at least 2 hours before it.
Storage and disposal
- Store at room temperature, 20-25°C (68-77°F).
- Do not freeze.
- Keep out of reach of children and pets.

Overdose
- If you think you have taken too many capsule, contact your doctor or go to the nearest emergency department straight away. Do not take additional doses unless your healthcare professional advises it.
- Since Betatinib capsule is taken at home, always keep it in its original container and never share it with anyone else.
Missed a Dose
- If you miss a dose of Betatinib capsule, take it as soon as you remember, as long as it is within 3 hours of your usual time. If more than 3 hours have passed, skip the missed dose and take your next capsule at the usual time.
- Do not take extra capsule to make up for a missed one. If you are not sure what to do, call your doctor or pharmacist for guidance.

Interactions
Interactions with other medicines
- Avoid strong antifungal medicines such as itraconazole and antibiotics such as clarithromycin, as they can raise the amount of Betatinib capsule in your blood and increase side effects; if you need one for a short time, your doctor will tell you whether to pause treatment.
- If you need a moderate-strength version of these types of medicines, your doctor will lower your dose of Betatinib capsule to once a day for that period.
- Avoid medicines such as rifampicin and carbamazepine (used for infections or seizures), as they can reduce the amount of Betatinib capsule in your blood and may reduce its effectiveness; if you cannot avoid them, your doctor will adjust your dose as appropriate.
- Do not take common stomach acid medicines called proton pump inhibitors (such as omeprazole and pantoprazole) during treatment, as they can reduce the amount of acalabrutinib absorbed into your body; ask your doctor for an alternative before starting.
- If you use an antacid or an H2-blocker for stomach acid, space it carefully from your dose of Betatinib capsule as your doctor instructs.
- Always tell your doctor about every medicine, supplement, vitamin, and herbal product you take before starting treatment.
Interactions with food items
- Betatinib capsule can be taken with or without food, as eating does not change how much medicine your body absorbs.
- Drink enough fluids, especially if you experience diarrhoea or vomiting.
- Ask your doctor whether alcohol should be limited or avoided based on your overall health and other medicines you take.
