• If you are allergic to dacomitinib or any other ingredient in this medicine, as a serious allergic reaction may occur.
• If you are pregnant, the active drug can cause severe harm or death to an unborn baby.
• If you are currently breastfeeding, it can develop potential adverse effects in nursing infants.

• Frequent loose or watery stools (diarrhoea)
• Skin reactions, including acne-like rash (acneiform dermatitis), dry skin, redness, and skin peeling
• Inflammation and infection around the nails (paronychia)
• Mouth sores, ulcers, and lip inflammation (stomatitis)
• Reduced appetite and weight loss
• Feeling sick (nausea) and vomiting
• Dry eyes, eye irritation, and conjunctivitis
• Hair loss or hair thinning (alopecia)
• Severe tiredness and physical weakness (fatigue)
• Raised liver enzymes on blood test panels

• Changes in taste (dysgeusia)
• Nosebleeds (epistaxis)
• Abnormal hair growth, such as excessive eyelash growth or brittle nails
• Dehydration resulting from persistent diarrhoea or nausea

• Severe Diarrhoea: Intense or persistent diarrhoea leading to severe dehydration, low blood pressure, and kidney injury, which requires prompt medical assessment and appropriate treatment.
• Interstitial Lung Disease (ILD) / Pneumonitis: Severe, potentially fatal lung inflammation; symptoms include new or worsening shortness of breath, cough, or fever. If ILD/pneumonitis is confirmed, treatment should be permanently discontinued.
• Severe Skin Reactions: Extensive rash, skin blistering, or skin peeling covering large areas of the body that may require treatment interruption or dose reduction.

• Take Dacoplice tablet exactly as your doctor tells you.
• You can take Dacoplice tablet with or without food.
• Swallow the tablets whole with a glass of water. Do not chew, crush, or split them.
• If you vomit or miss a dose, do not take an extra tablet; take your next dose at the usual scheduled time the following day.

• Store tablets at the storage temperature specified on the product packaging/prescribing information.
• Keep the tablets in their original packaging to protect them from moisture and light.
• Keep out of reach of children and pets.
• Dispose of unused/expired medicines properly.

• Keep anti-diarrheal medication (such as loperamide)available if advised by your doctor, and follow the recommended treatment for diarrhoea.
• Apply alcohol-free moisturizing creams regularly to your skin and hands to manage dry skin and prevent acne-like skin rashes.
• Avoid direct sunlight and wear protective clothing and sunscreen outdoors, as Dacoplice tablet can make your skin more sensitive to sunlight.
• Practice good oral hygiene by using a soft toothbrush and alcohol-free mouthwashes to help prevent painful mouth sores.
• Contact your doctor immediately if you develop new or worsening shortness of breath, cough, or chest pain, as these may signal severe lung inflammation.
• Women who could become pregnant must use effective birth control during treatment and for the period recommended in the prescribing information after the last dose.

Interactions with other medicines

• Strong CYP2D6 substrates with narrow therapeutic indices (such as procainamide, flecainide, or certain tricyclic antidepressants) may have increased blood levels when taken with Dacoplice tablet, potentially increasing the risk of adverse effects.
• Strong CYP3A4 inducers (such as rifampicin, carbamazepine, phenytoin, and St. John's Wort) can significantly lower the concentration of Dacoplice tablet in your blood and reduce its effectiveness.
• Proton pump inhibitors (PPIs, like omeprazole or pantoprazole) can reduce the absorption of Dacoplice tablet. If acid-reducing therapy is required, an antacid or H2-receptor antagonist may be considered according to your doctor's advice.
• Always tell your doctor about all prescription drugs, over-the-counter medicines, vitamins, and herbal supplements you take before starting treatment.
