Description
Estadep Plus tablet is used to treat comorbid depression and anxiety disorders. It contains clonazepam and escitalopram as active ingredients. Estadep Plus Tablet helps relax mood by lowering the exc
ess activity of nerves in the brain. It also has sedative effects that relieve anxiety and improve sleep. Depression is a mood disorder characterised by episodes of sadness, loss of interest, mood swings, hopelessness, etc. In contrast, anxiety is characterised by episodes of excessive, unrealistic worry and fear that can interfere with daily activities. Estadep Plus tablet should only be taken after a doctor’s consultation. The dose of the Estadep Plus tablet should be carefully monitored, as there are many side effects associated with this medicine, especially in older patients and persons with liver and kidney disorders.

Uses
- As per CDSCO, the Estadep Plus tablet is used for the following cases:
- Treatment of comorbid depression (depression associated with other disorders)
- Treatment of anxiety disorder
Contraindications
- Estadep Plus Tablet is contraindicated in the following cases:
- If you are allergic to any active component of Estadep Plus tablet, i.e. Clonazepam, Escitalopram
- If you are already taking Monoamine Oxidase Inhibitors (MAOIs): linezolid, methylene blue, Selegiline, antipsychotic medicine-pimozide, anti-anxiety or sedative medications
- If you are diagnosed with acute angle glaucoma (an eye condition that increases the fluid pressure in the eyes)
- If you have a seizure disorder and taking antiepileptic (anti-seizure) medications
- If you have liver or kidney disease
- If you have respiratory issues
- If you have a severe heart condition
- If you are pregnant or breastfeeding

Mode of Action
How Does It Work?
Estadep Plus Tablet contains two active ingredients: Clonazepam and Escitalopram. Clonazepam is a benzodiazepine that acts as a GABA-A (Gamma Amino Butyric Acid) receptor agonist. GABA is an inhibitory neurotransmitter. By binding to the GABA-A receptor, it enhances the inhibitory effects of GABA, resulting in sedative, anti-anxiety and muscle relaxant properties and thus, relieving anxiety and depression symptoms.
Escitalopram is an antidepressant belonging to the Selective Serotonin Reuptake Inhibitors (SSRIs) class of medicine. It works by preventing the reuptake of a chemical messenger called serotonin by the nerve cells, resulting in an increased serotonin level in the brain. Serotonin is a natural chemical messenger responsible for controlling behavioural processes such as mood, fear, anger, appetite, memory, etc....
Directions for Use
- Take the Estadep Plus tablet as per the doctor’s prescription.
- Take the medicine with or without food or as prescribed. In the case of any stomach illness, take it after meals.
- Take Estadep Plus tablet preferably at the same time every day to maintain its constant level in the body.
- Do not break/crush the tablet or chew this medicine; take it as a whole with a glass of water.
- Do not take Estadep Plus tablet with other medications; consult your doctor first.
- Do not stop taking this medication in between, as it may lead to withdrawal side effects.
- Do not exceed the dosage on your own without consulting a doctor.
Interactions
Interactions with other medicines
- Many medicines can interact with the Estadep Plus tablet, making it less effective or increasing side effects.
- Estadep Plus tablet should not be used concomitantly or within two weeks of using Monoamine Oxidase Inhibitors (MAOIs) like isocarboxazid, linezolid, methylene blue, selegiline, rasagiline, etc. It is potentially life-threatening....
- Concomitant use of Estadep Plus tablet with antidepressants like Clomipramine, Desipramine, Imipramine, Amitriptyline, Amoxapine, etc.; opioids and other central nervous system depressants is contraindicated, as it may cause respiratory depression, coma and death....
- Estadep Plus tablet interaction with metoprolol can lead to excessive bradycardia (slow heart rate).
- Estadep Plus tablet interaction with antipsychotics (pimozide, haloperidol), tricyclic antidepressants and antihistamines can prolong QT interval on ECG and may alter heart rhythm.
- Estadep Plus tablet interaction with NSAIDs, aspirin, and other medications that affect coagulation can increase the risk of bleeding and bruises.
- Estadep Plus tablet may also interact with herbal medicine like St John’s wort and lead to life-threatening side effects.
Interactions with food items
- Avoid consuming Estadep Plus tablets with cola drinks, chocolates, and caffeine-containing drinks like tea and coffee. It can result in serotonin syndrome (characterised by high blood pressure, confusion, and muscle rigidity). It would help if you also avoided tyramine-rich beverages like cheese, milk, beef, chicken liver, meat extract, soybeans and grapefruit....
Storage and disposal
Store Estadep Plus tablet away from sunlight. Keep it at room temperature in a cool and dry area. Keep it away from children and pets. Keep the medicine within its original packaging.
Do not flush Estadep Plus tablets down into the toilet or throw them into the dustbin unless instructed to do so. Medicines disposed of in this manner may contaminate the environment. Properly dispose of this product when it is expired or no longer required....
Dosage
Overdose
Estadep Plus tablet overdose can lead to side effects like irregular heartbeat, low blood pressure, slowing of heart rate, sleepiness, double vision, difficulty breathing, cardiac arrest, ECG changes (change in heart functioning) and coma. If you experience any of these symptoms or have taken an overdose of this medicine, immediately seek emergency medical treatment from a nearby hospital....
Missed a Dose
If you missed a dose of your Estadep Plus tablet, take it as soon as you remember. If you recall missing a dose when it is almost the time to take the next dose, skip the missed one and continue with your regular dosing schedule. Do not use them together, as this can lead to an overdose....
