Everolimus

• Everolimus is used in the management of
• Neuroendocrine Tumors of the Pancreas, gastrointestinal tract, or lung (not indicated for functional carcinoid tumors)
• Advanced Renal Cell Carcinoma (RCC), after failure of prior treatment with a VEGF-targeted therapy
• Tumor associated with (Subependymal giant cell astrocytoma) tuberous sclerosis and renal angiomyolipoma.
• Seizures associated with tuberous sclerosis complex (as an add-on treatment)
• Prophylaxis to prevent rejection of organs in patients with kidney or liver transplant
• Note: Everolimus is not recommended for the prevention of rejection in heart transplant patients in this labeling, due to a documented increase in mortality (often linked to serious infection) within the first three months after transplant.

When should one not use Everolimus

• Everolimus is contraindicated in patients with a known allergy/hypersensitivity to everolimus.

Common side effects of this drug are:

• Very Common
• Mouth sores or ulcers (stomatitis)
• Infections, Rash
• Tiredness and weakness
• Diarrhoea, nausea
• Swelling of the arms or legs
• Fever
• Reduced appetite
• Common
• Cough, shortness of breath
• Headache
• Altered taste
• Nosebleeds
• Raised blood sugar, cholesterol, or triglycerides
• Low blood cell counts (anaemia, low white cells, low platelets)
• Difficulty sleeping
• Serious
• New or worsening cough, shortness of breath, or chest tightness
• Signs of serious infection like fever, chills, unusual tiredness
• Severe allergic reaction with difficulty breathing, swelling of the face, lips, tongue, or throat
• Reduced urination or impaired kidney function
• Unusual bleeding, bruising, or infection due to low blood cell counts
• Uncontrolled high blood sugar or cholesterol

Other General Warnings

• Consult your doctor:
• If you have any lung condition or breathing problems.
• If you have a history of kidney disease.
• If you have diabetes or high cholesterol.
• If you are due to have any live vaccinations
• If you have any planned or recent surgery.
• Report new or worsening cough, breathlessness, fever, or mouth sores to your doctor promptly.
• Attend all scheduled blood tests to monitor blood counts, kidney function, blood sugar, and cholesterol.
• Avoid live vaccines and close contact with recently vaccinated individuals while on treatment.
• Tell your doctor about all other medicines and supplements you take.
• Keep out of the sight and reach of children and pets at all times.

How Does It Work?

Interactions with other medicines

• Some antifungal (like ketoconazole or itraconazole) and antibiotic medicines (like clarithromycin or erythromycin) can raise Everolimus levels and should generally be avoided; if unavoidable, your doctor will adjust your dose.
• Seizure medicines (like carbamazepine, phenobarbital ) and other enzyme-inducing medicines can lower Everolimus levels and reduce its effectiveness; your doctor may need to increase your dose.
• Tell your doctor if you take blood pressure medicines (ACE inhibitors), as combining them with Everolimus may increase the risk of facial or throat swelling.
• Always tell your doctor about all prescription medicines, over-the-counter medicines, vitamins, supplements, and herbal products you take.

Missed a Dose
