How Does It Work?
Primidone is an anticonvulsant; it works by decreasing the abnormal electrical activity in the brain.
Directions for Use of Prolet 12.5 MG
- Take this tablet for the dose and duration prescribed by your physician
- Do not break, chew or crush this tablet
- Swallow the tablet with a glass of water
Interactions of Prolet 12.5 MG
Interactions with other medicines
- Antiviral medicines, anti-infectious agents and antifungal agents like voriconazole may have lower plasma concentrations as a result of accelerated metabolism caused by primidone
- Co-administration of pyrimidone with St John's Wort reduces its efficacy and decreases the plasma concentration of primidone
- For drugs the following drugs, there is a risk of lower plasma concentrations due to accelerated metabolism caused by primidone and dose adjustments might be needed:
- Drugs affecting the nervous system: oxycodone
- Anticoagulant drugs: dabigatran, ticagrelor
- Anti-neoplastic agents: tyrosine kinase inhibitors
- Cardiovascular agents: bosentan, nimodipine
- Hormonal agents: abiraterone, ulipristal
- Drugs affecting the nervous system when co-administered with primidone may also have an increased risk of additive CNS depression

Missed a Dose
If you forget to take a dose, take it as soon as you remember. If the next dose is approaching, skip the missing dose. Instead, use your next dose at the same time every day. Do not take two tablets at once....

Q: I am a patient with renal insufficiency, is there anything I should know before starting the treatment?
A: Conditions like renal insufficiency decrease the renal elimination of primidone in patients; therefore, dose adjustments are needed according to biological monitoring and clinical response. Make sure you inform the doctor about your condition and your current medications.
Q: Can I stop this medication when I feel better?
A: You should not stop this medication as sudden withdrawal may induce convulsive fits. Instead, please continue the entire duration of the treatment and if you are experiencing side effects and want to stop the medication, talk to your doctor about it.
