Mode of Action

How Does It Work?

Lorazepam is a benzodiazepine that acts on the brain pathways to increase the action of a chemical messenger called GABA (gamma-aminobutyric acid) and increases its activity. This results in sedation, calming effects and muscle relaxation. These effects are beneficial in the treatment of anxiety. It is a sedative, anxiolytic, and hypnotic agent. The advantage of Lorazepam is in its rapid onset of action (within 1-3 minutes), which proves useful in acutely managing situations of sudden, increased anxiety.

  • Clozapine and Lorazepam should not be used together, as they may produce marked sedation, excessive salivation, drop in blood pressure, lack of muscle control, delirium and respiratory arrest.
  • The concomitant use of Lorazepam and Valproate is not recommended as it increases the concentration of Lorazepam in blood and causes side effects.
  • Probenecid prolongs the duration of action of Lorazepam.
  • Theophylline and aminophylline decrease the sedative effects of Lorazepam and other benzodiazepines.
