Medical Description
Jakavi tablet contains ruxolitinib, a JAK inhibitor. It is used to treat myelofibrosis, polycythaemia vera, and certain cases of acute graft-versus-host disease (GvHD) when previous treatment has not worked well enough.
It works by blocking JAK1 and JAK2 proteins, which are involved in blood-cell production and immune responses. This helps reduce abnormal blood-cell activity and inflammation.
Common side effects include anaemia, low platelet or white blood cell counts, infections, headache, dizziness, and increased cholesterol levels. Regular blood tests are needed to monitor your blood counts, and your doctor may also check your liver, kidney, and cholesterol levels.
Tell your doctor if you have liver or kidney problems, an infection, hepatitis B, or tuberculosis, or if you take other medicines, supplements, or herbal products.
Uses
- Myelofibrosis, including primary myelofibrosis, post-polycythemia vera myelofibrosis, and post-essential thrombocythemia myelofibrosis.
- Polycythemia vera in patients who are resistant to or cannot tolerate hydroxyurea.
- Acute graft-versus-host disease (GvHD) in adults and children aged 12 years and older who have been previously treated unsuccessfully with steroid medications.

- Increased liver enzymes
- Increased blood creatinine
- Increased creatine phosphokinase (CPK)
- Serious infections, including tuberculosis, hepatitis B reactivation, herpes zoster, and sepsis
- Serious bleeding, particularly when blood platelet counts are low
- Thrombosis, including venous thromboembolism
- Progressive multifocal leukoencephalopathy (PML), a rare serious brain infection
- Non-melanoma skin cancers
- Major cardiovascular events, particularly in patients with cardiovascular risk factors
- Severe allergic reactions
Precautions and Warnings
Other General Warnings
- You have low blood cell counts or develop signs of infection.
- You have a serious infection, tuberculosis, or hepatitis B.
- You develop symptoms of shingles.
- You develop new memory, neurological, or mental health changes.
- You have high cholesterol or triglyceride levels.
- You are 65 years or older, or are a current or former long-term smoker.
- You have heart disease or risk factors for blood clots.
- You develop leg swelling or pain, chest pain, or shortness of breath.
- You have a history of skin cancer or precancerous skin lesions; regular skin checks may be recommended.
- You have kidney or liver problems, as your dose may need adjustment.
- Your symptoms return or worsen after stopping or interrupting Jakavi tablet.
Directions for Use
- Jakavi tablet should be taken orally, with or without food, as prescribed by your doctor.
- Take the medicine at approximately the same time each day. Do not change the dose or stop treatment without consulting your doctor.
Storage and disposal
- Store the medicine as directed on the product label.
- Keep Jakavi tablet out of the reach of children.

Missed a Dose
- If you miss a dose, skip it and take the next usual dose at the scheduled time.
- Do not take an extra dose to make up for the missed dose

Interactions
Interactions with other medicines
- CYP3A4 inhibitors: Medicines such as ketoconazole, clarithromycin, and erythromycin may increase ruxolitinib levels and the risk of side effects. Your doctor may adjust the dose.
- CYP3A4 inducers: Medicines such as rifampicin, carbamazepine, and phenytoin may lower ruxolitinib levels and reduce its effectiveness.
- Tell your doctor if you take antibiotics or antifungal medicines, as some may interact with ruxolitinib.
- Always inform your doctor about all medicines, supplements, and herbal products you take.
