Molecule Description
Tadalafil+Ambrisentan is a combination medication that contains ambrisentan and tadalafil as its main ingredients. It is prescribed for pulmonary arterial hypertension (PAH), a condition characterized by high blood pressure in the arteries transporting blood from the heart to the lungs. It helps improve quality of life and exercise capacity while also delaying disease progression. Ambrisentan is an endothelin receptor blocker and tadalafil is a phosphodiesterase type 5 (PDE5) inhibitor; together they relax and widen the blood vessels in the lungs, which reduces blood pressure in the pulmonary arteries.
It is contraindicated in patients with severe liver disease or a known allergy to the medication. Common side effects may include peripheral oedema, nasal congestion, sinusitis, and flushing. Dizziness can also occur, which may affect activities requiring mental focus. Take this medication as prescribed by your doctor, preferably at the same time each day, with or without food.
Consult your doctor about all current medications, as Tadalafil+Ambrisentan may interact with other drugs. Potential interactions with other medications should be assessed by a healthcare provider. While no specific food interactions are known, consult your doctor for tailored advice.

Mode of Action
How Does It Work?
Tadalafil+Ambrisentan combines two medicines. Ambrisentan is an endothelin receptor blocker; it inhibits endothelin, a natural compound that constricts blood vessels. Tadalafil is a phosphodiesterase type 5 (PDE5) inhibitor that relaxes and widens the blood vessels in the lungs. Together they relax the pulmonary arteries, facilitate blood flow, reduce the heart's workload and lower pulmonary arterial pressure, which alleviates symptoms.
Interactions
Interactions with other medicines
- To avoid any possible interaction, tell your doctor about all the medicines, supplements, or herbal supplements you are currently taking or might take.
- Especially if you are taking any other medicine to treat PAH.
- Other medicines that should be used with caution are immunosuppressants like ciclosporin and anti-tuberculosis medicines like rifampicin.
- Concomitant administration of Tadalafil+Ambrisentan along with medicines containing nitrates leads to severe hypotensive effects (low blood pressure).
