Medical Description
Vault tablet is used for the treatment and prevention of sores in the lining of the stomach or intestine. It contains vonoprazan as the main ingredient, which lowers the amount of acid the stomach makes.
Vonoprazan is a medication that reduces stomach acid by blocking the proton pump, an enzyme in the stomach cells. This process halts the final step of acid production. Unlike other medications, vonoprazan does not require stomach acid to function and can accumulate in the stomach cells regardless of their activity. It temporarily binds to the proton pumps, allowing them to work effectively.
Swallow this medicine whole with plenty of water. You may take it with or without food. Common side effects of Vault tablet include diarrhoea, stomach pain, nausea, high blood pressure, and indigestion. If you experience any of these symptoms, it is essential to consult your healthcare provider.
Uses
- Vault tablet is used to heal damage to the esophagus caused by excess stomach acid (erosive esophagitis) and alleviate associated heartburn.
- It also helps maintain healing and prevent recurrence of heartburn after the esophagus has healed. When prescribed with other antibiotics (such as amoxicillin alone or amoxicillin and clarithromycin), it helps eliminate Helicobacter pylori bacteria.
Contraindications
- Do not take Vault tablet if:
- You are allergic to vonoprazan or any of the other ingredients of this medicine. Allergic reactions to vonoprazan have included severe reactions such as anaphylaxis.
- You are taking rilpivirine-containing medicines (used to treat HIV). Because Vault tablet reduces stomach acid, it can make rilpivirine less effective.

- Headache
- Altered sense of taste (more likely when taken with antibiotics for H. pylori)
- Vaginal yeast infection (thrush) or urinary tract infection (when used with antibiotics for H. pylori)
- Severe skin reactions (spreading rash, blistering, peeling skin, or mouth sores)
- Serious allergic reactions (swelling of the face, lips, or throat, or difficulty breathing)
- Persistent or severe watery diarrhoea
- Signs of kidney problems
- Signs of low magnesium (muscle cramps, tremors, or an irregular heartbeat)

Directions for Use
- Take Vault tablet exactly as prescribed by your doctor. Swallow the tablet whole with enough water. Do not chew or crush it. It can be taken with or without food. Vonoprazan is available as 10 mg and 20 mg tablets; your doctor will decide the strength, dose, and duration based on your condition.
- Erosive esophagitis is usually treated for several weeks, sometimes followed by a lower maintenance dose, while Helicobacter pylori infection is treated together with antibiotics for a shorter, fixed course. Complete the full course even if you feel better.

- You should inform your doctor if you have a low level of magnesium or any other mineral, or if you have low vitamin B12 levels. If you are undergoing long-term treatment (more than one year) with this medication, it may decrease magnesium levels or lead to bone weakness. Consult your doctor to determine if you may need supplements to address these deficiencies.
- Additionally, inform your doctor if you experience any issues with your bones, kidneys, or liver.
- You can make certain lifestyle modifications, such as avoiding excessive intake of acidic foods (such as carbonated drinks and oily or fried foods), engaging in regular exercise, and maintaining a healthy weight.
- It is also advisable to limit alcohol intake and quit smoking to improve the treatment of your acid reflux.

Interactions with other medicines
- Some medicines can affect Vault tablet, which can affect how they work. Tell your doctor about all the medicines, supplements, and herbal products you take. In particular:
- Rilpivirine (HIV medicine) - must not be taken with this medicine.
- Atazanavir, nelfinavir, and other HIV medicines - Vault tablet can change how well they are absorbed; your doctor may avoid combining them.
- Medicines that need stomach acid to be absorbed, such as iron salts, erlotinib, dasatinib, nilotinib, mycophenolate mofetil, and ketoconazole/itraconazole - Vault tablet may reduce their effect.
- Clopidogrel (used to prevent blood clots) - Vault tablet may reduce how well clopidogrel works, so your doctor may monitor you or suggest an alternative.
- Citalopram (an antidepressant) and cilostazol - their levels may rise, so your doctor may watch for side effects.
- Digoxin and water pills (diuretics) - relevant if your magnesium level drops (see warnings).
- Medicines that speed up drug breakdown in the liver (strong CYP3A4 inducers) - these can make Vault tablet less effective and are usually avoided.
- Tell your doctor, especially if you are taking medicines for HIV/AIDS.
