- You have a history of seizures or fits.

- You have difficulty passing urine, a history of urinary retention, or prostate problems.
- You have glaucoma, especially narrow-angle glaucoma, or increased pressure in the eyes, as even normal doses may trigger an attack.
- You have any heart problems, such as irregular heartbeat, fast heart rate, heart failure, or a previous heart attack.
- You have ever had a stroke or other blood circulation problems.
- You have thyroid problems or are taking thyroid medicines.
- You are taking any other antidepressants, as combining them may increase side effects without added benefit.
- You are scheduled for surgery, as this medicine may need to be stopped a few days before the procedure.
Directions for Use
- Amitop M Tablet should be taken as directed by your doctor.
- Swallow it whole with a glass of water.
- It should be taken with or after a meal to reduce its side effects.
- It is best to take it at a fixed time for optimal results, and you should not consume it more than prescribed by your doctor.

How Does It Work?
- Vitamin B12 (Mecobalamin / Cyanocobalamin / Methylcobalamin): Vitamin B12 helps keep nerves healthy and supports the formation of red blood cells. It plays an important role in repairing damaged nerves, improving nerve signal transmission, and reducing symptoms like numbness, tingling, and weakness. It also helps prevent and treat vitamin B12 deficiency–related anemia and supports overall brain and nerve function.
- Amitriptyline: Amitriptyline works by increasing the levels of certain natural chemicals (such as serotonin and norepinephrine) in the brain that help improve mood and reduce pain signals. At low doses, it is commonly used to relieve nerve pain, improve sleep, and reduce symptoms like anxiety or chronic pain by calming overactive nerve pathways.
Interactions
Interactions with other medicines
- Amitriptyline may reduce the blood-pressure-lowering effect of medicines like guanethidine.
- When taken with medicines that have anticholinergic effects (such as some allergy or bladder medicines) or drugs that stimulate the nervous system (including adrenaline used with local anesthetics), close monitoring is needed as side effects may increase.
- Using amitriptyline together with anticholinergic medicines can cause severe constipation or bowel blockage, especially in elderly or hospitalized patients.
- If constipation occurs while taking amitriptyline with such medicines, it should be managed promptly to avoid serious bowel problems.
- Cimetidine (a medicine for acidity) may slow the breakdown of amitriptyline in the liver, increasing its effects and side effects.
- Taking large doses of ethchlorvynol with amitriptyline may cause confusion or delirium.
- Amitriptyline can increase the effects of alcohol, sleeping pills, and other medicines that slow the brain, leading to excessive drowsiness or confusion.
- Confusion has been reported when amitriptyline is taken together with disulfiram (used in alcohol dependence).
- High fever has been reported when amitriptyline is used with anticholinergic or antipsychotic medicines, especially in hot weather.
- Absorption of vitamin B12 may be reduced when taken with medicines like neomycin, aminosalicylic acid, H2 blockers (for acidity), or colchicine.
- Oral contraceptive pills may reduce vitamin B12 levels in the blood.
- The effect of vitamin B12 in treating anemia may be reduced when chloramphenicol injections are used at the same time.
