Molecule Description
Vardenafil is a medicine used to help men who have difficulty getting or maintaining an erection. It improves blood flow to the penis, but it works only when you are sexually stimulated. This means the medicine does not cause an erection on its ownyou still need sexual arousal for it to work effectively. It is usually taken with a glass of water, and following your doctor's instructions helps you get the best results.
You should take this medicine exactly as prescribed. It can be taken with or after meals, which may help reduce stomach-related side effects. However, very heavy or high-fat meals may delay how quickly it starts working. Take it only when needed, and do not take more than one dose in 24 hours. Avoid taking more than the prescribed dose, as this increases the risk of side effects such as headache, dizziness, or nasal congestion.
Some men may experience dizziness or visual changes after taking this medicine, so it's important to see how your body responds before driving or operating machines. Alcohol can be taken with Vardenafil, but drinking too much alcohol may make it harder to achieve an erection. Grapefruit or grapefruit juice should be avoided because it can increase the level of the medicine in your body and raise the chances of side effects.
This medicine interacts with several other drugs. It should never be taken with nitrates, nitric-oxide donors, nicorandil, or riociguat, as these combinations can cause a dangerous drop in blood pressure. Strong HIV medicines, antifungal medicines like ketoconazole or itraconazole, and antibiotics such as erythromycin or clarithromycin can also increase its levels in the body. If you are taking alpha-blockers for prostate or blood pressure problems, your doctor may start you on a lower dose to prevent dizziness or low blood pressure.
Before taking Vardenafil, inform your doctor if you have heart problems, very low blood pressure, severe liver disease, bleeding issues, or eye conditions such as sudden vision loss. This medicine is not meant for women and should not be used during pregnancy or breastfeeding. If you ever experience sudden vision changes, chest pain, or an erection lasting more than 4 hours, stop using the medicine and seek medical help immediately.

- You are taking alpha-blockers, as the combination may cause dizziness or low blood pressure and may need dose adjustments.
- You are taking strong medicines that affect liver enzymes (such as ketoconazole, itraconazole, erythromycin, or clarithromycin), so your dose may need to be adjusted or the combination avoided.

Mode of Action
How Does It Work?
Vardenafil works by relaxing the blood vessels in the penis, which improves blood flow when you are sexually stimulated. It blocks an enzyme called PDE5, which normally limits blood flow, allowing a stronger, longer-lasting erection. It does not cause an erection on its own; sexual arousal is still needed for it to work.
Interactions
Interactions with other medicines
- Strong HIV medicines like indinavir and ritonavir can greatly increase vardenafil levels in the body, making side effects more likely.
- Strong antifungal medicines like ketoconazole and itraconazole can raise vardenafil levels and should not be used together.
- Antibiotics such as erythromycin and clarithromycin can increase vardenafil levels so that a lower dose may be needed.
- Medicines such as ranitidine, digoxin, warfarin, glibenclamide (diabetes medicines), ACE inhibitors, beta-blockers, and diuretics do not significantly affect vardenafil.
- Alcohol does not increase vardenafil's side effects but may reduce sexual performance on its own.
- Taking nitrates or nitric oxide–releasing medicines with vardenafil can cause a dangerous drop in blood pressure and must be avoided.
- Nicorandil (contains nitrate) can interact seriously with vardenafil and should not be taken together.
- Taking vardenafil with alpha-blockers (for prostate problems or blood pressure) may cause dizziness or low blood pressure; start vardenafil at a low dose and separate doses if needed.
- Riociguat should not be taken with vardenafil because it can cause severe low blood pressure.
Interactions with food items
- Vardenafil can be taken with or without food, but a very heavy or high-fat meal may delay its onset.
- Grapefruit juice can increase vardenafil levels in the body, potentially increasing the risk of side effects, so it is best to avoid it.
