- Major depressive disorder (MDD)

Contraindications
- Hypersensitivity reactions.
- Recent history of arrhythmias, myocardial infarction, heart failure, or QTc prolongation.
- Angle closure glaucoma.
- Epilepsy/seizure disorders patients.
- Pulmonary diseases.

Directions for Use
- This medication should be taken as prescribed by the treating physician.
- Unexpected stoppage of the medication can compel withdrawal side effects.
- Consult your doctor before stopping this medication.
- It is recommended to consume this medication on a regular schedule, i.e., at the same time every day, for optimal pharmacological effects of this medication.
- Swallow this medication as a whole. Do not granulate, break or bite this tablet.
- Inform your doctor regarding other medications you may be taking to review and prevent any medication interaction associated effects.

Overdose
- Amitriptyline harmfulness is quantifiable by a portion of more than 5 mg/kg. Amitriptyline toxicity can be severe and even lethal., requiring ICU admission and intervention. In the event of overdose, the patient may have sleepiness, seizures, coma, raised heart rate, low blood pressure, irregular heart rate, dilated pupils, dry mouth, decreased (or absent) bowel sounds, inability to pass urine. In case of a suspected overdose, immediately rush the patient to the nearest tertiary hospital.
- In cases of chlordiazepoxide overdose or toxicity, the patient may show signs of confusion, slow reflexes, drowsiness, or coma. This requires immediate intervention, and the patient should be rushed to a hospital immediately.

Interactions with other medicines
- This medication causes prolongation of QTc interval and should not be used in addition to other medicines that also have the same effect. It may result in cardiac arrhythmias and other potentially life-threatening conditions. Examples include astemizole, cisapride, disopyramide, ibutilide, indapamide, pentamidine, pimozide, procainamide, quinidine, sotalol, terfenadine.
- Amitriptyline gets utilised through CYP3A4. A few medications modify the action of CYP3A4. The dose should be mindfully controlled, just as the whole understanding medication routine checked for CYP3A4 inducers and inhibitors.
- It is recommended to discontinue this medication before elective surgery to decrease the risk of interaction with anaesthetic agents.
- This medication should not be used:
- Concomitantly with MAOIs (Monoamine oxidase inhibitors) or within 14 days of the use of MAOIs and vice versa. Examples of MAOIs include Selegiline, rasagiline.
- With other medications known to increase serotonin levels. For example, isocarboxazid, phenelzine, procarbazine, safinamide, selegiline, tranylcypromine, sertraline as it may cause Serotonin syndrome, a potentially fatal condition.
- With opioid medications, benzodiazepines, or CNS depressants.
