A quick info on ANA test
Overview
The antinuclear antibody (ANA Elisa) test is a blood test that detects antinuclear antibodies (ANA), which may be present in people with certain autoimmune conditions. Antibodies are proteins made by the immune system to help protect the body from infections and other harmful substances. Normally, they identify and attack foreign substances such as bacteria and viruses. Sometimes, however, the immune system mistakenly produces antibodies that target the body’s own healthy cells. These are called autoantibodies. Antinuclear antibodies are a type of autoantibody that targets substances found inside the nucleus of cells. The presence of ANA in the blood may be associated with autoimmune diseases such as systemic lupus erythematosus (SLE), Sjögren syndrome and systemic sclerosis. However, ANA can also be found in some healthy people and in certain infections or other conditions.
Doctors may recommend the test when symptoms such as persistent tiredness, joint or muscle pain, swelling, skin rashes, fever, and unexplained changes in overall health are present, which may suggest an autoimmune disorder. Symptoms can vary depending on the condition and the organs affected. A positive ANA result does not confirm an autoimmune disease on its own. Doctors interpret the result along with symptoms, medical history, and other test findings.
When and Who Should Get an ANA-Elisa Test Done?
The ANA Elisa test is generally considered when your symptoms cannot be explained by other conditions or when an autoimmune disease is suspected based on your medical history and examination. Your doctor may recommend this test when there are signs or symptoms such as:
- Persistent or unexplained fatigue
- Joint pain, stiffness, or swelling
- Muscle pain or weakness
- Recurrent fever
- Unexplained skin rashes, blisters, or changes in skin colour
- Swelling of the salivary glands, particularly with dry mouth or dry eyes
- Symptoms affecting multiple parts of the body
- Unexplained abdominal pain
- Unexplained hair loss
- Increased sensitivity to sunlight
- Fingers or toes that turn unusually pale, blue, or red, especially in cold weather
Note: ANA testing is not usually recommended as a routine screening test for people without symptoms suggestive of an autoimmune disorder. This is because antinuclear antibodies can also be found in some healthy people, so a positive result does not necessarily mean that you have an autoimmune disease.
What Conditions Can an ANA-Elisa Test Help Detect?
The ANA-Elisa test can help doctors evaluate certain autoimmune and connective tissue diseases. ANA is commonly associated with conditions such as:
- Systemic lupus erythematosus (SLE): A long-term autoimmune disease that can affect several parts of the body, including the joints, skin, kidneys, heart, lungs, blood vessels, and nervous system.
- Sjögren syndrome: An autoimmune condition that commonly affects the glands that produce tears and saliva, causing dry eyes and dry mouth.
- Scleroderma (systemic sclerosis): An autoimmune condition that can cause changes in the skin and may affect blood vessels and internal organs.
- Inflammatory muscle diseases (myositis): A group of conditions that can cause inflammation and weakness of the muscles.
- Rheumatoid arthritis: An autoimmune condition that mainly affects the joints, causing pain, stiffness, and swelling.
- Psoriatic arthritis: An inflammatory condition that can cause joint pain, stiffness, and swelling in people with psoriasis.
- Autoimmune hepatitis: A condition in which the immune system attacks the liver, causing inflammation.
- Autoimmune thyroid diseases: Conditions in which the immune system attacks the thyroid gland and may affect thyroid hormone production.
Note: ANA is not specific to any one disease and cannot diagnose these conditions on its own. A positive ANA result may also occur in some healthy people and in certain other conditions. Doctors interpret the result along with your symptoms, medical history, examination, and other test results.
ANA-Elisa Test Preparation
What to Expect Before the ANA-Elisa Test?
- No special preparation is usually needed before an ANA test. You can eat and drink normally, as fasting or dietary changes are generally not required.
- The ANA test can be performed at any time of day.
- It is important to let your doctor know about medicines and supplements you take, as some medications may affect ANA-Elisa levels. Also, let them know if you had any viral illnesses recently.
What to Expect During the Blood Collection?
- A trained phlebotomist will visit your home at the scheduled time for sample collection.
- You will be seated comfortably while a suitable vein, usually near the inner elbow, is identified and cleaned with an antiseptic.
- A sterile needle will be used to collect a small blood sample, and you may feel a brief pinching sensation.
- Once the sample is collected, the needle will be removed, gentle pressure will be applied, and a small bandage will be placed over the site.
- The entire procedure is quick and typically takes only a few minutes.
What to Expect After the ANA-Elisa Test?
- You may feel a mild throbbing pain, which will pass in a few minutes. The sample will be sent to a lab to be analysed. Your test results should be available for download in a day or two on the PharmEasy platform.
- If your ANA test is positive, your doctor may recommend additional tests based on your symptoms and medical history to evaluate for a specific autoimmune condition.
Note: If you notice anything unusual at the puncture site, such as persistent bleeding, a rash, or pain, consult your healthcare provider.
What Are the Parameters and their Ranges Included in the ANA-Elisa Test?
The ANA-Elisa test measures antinuclear antibodies (ANA) in your blood using the enzyme-linked immunosorbent assay (ELISA) method. The result is reported as an OD (optical density) ratio.
|
ANA result |
OD ratio |
|---|---|
|
Negative |
< 0.90 |
|
Equivocal |
0.90–1.10 |

The reference range and cut-off values may vary depending on the laboratory and the testing method used. Your ANA-Elisa report will include the applicable reference range for interpreting your result.
ANA-Elisa Test Interpretation
|
ANA Result |
Interpretation |
|---|---|
|
Negative |
ANA is not detected at a significant level. However, a negative result does not completely rule out an autoimmune disease, particularly in the early stages or when other specific autoantibodies are involved. |
|
Equivocal |
The result is borderline. Your doctor may recommend repeat testing or additional tests for specific antibodies, depending on your symptoms and medical history. |
|
Positive |
ANA is detected above the laboratory’s cut-off. ANA positivity is commonly associated with conditions such as systemic lupus erythematosus (SLE), systemic sclerosis, Sjögren’s syndrome and rheumatoid arthritis. Your doctor may recommend tests for specific autoantibodies, such as anti-Sm, anti-RNP, anti-Ro or anti-La, to further evaluate a suspected autoimmune condition. |
A positive ANA result is not specific to any one condition. ANA may also be found in some healthy individuals, particularly older adults, as well as in certain infections, malignancies, and drug-induced autoimmune conditions. The final diagnosis is made by a doctor based on the overall clinical picture, and other relevant investigations.
ANA-Elisa Test Risks and Limitations
Potential Risks and Complications
There is minimal risk involved with having blood taken for the ANA test. Blood vessels may vary in size from one person to another and from one side of the body to the other. Taking blood from some people may be more difficult than from others.
Other risks associated with having blood drawn are slight but may include:
- Fainting or feeling light-headed
- Multiple punctures to locate veins
- Bleeding
- Infection
Understanding ANA-Elisa Test Limitations
- A positive ANA result does not confirm an autoimmune disease or identify a specific condition on its own.
- ANA may be detected in healthy individuals, particularly older adults, as well as in some people with infections, malignancies or drug-induced autoimmunity.
- A negative ANA result does not completely rule out an autoimmune disease, particularly in the early stages of some conditions or when other specific autoantibodies are involved.
- ANA testing does not indicate which autoimmune disease is present. Additional antibody tests and other investigations may be needed based on your symptoms and medical history.
- Certain factors, such as problems with the blood sample or some medicines and substances such as high-dose biotin, may interfere with the test and affect the accuracy of the result.
